Vaccine adjuvant uses host DNA to boost pathogen recognition

Aluminum salts, or alum, have been injected into billions of people as an adjuvant to make vaccines more effective. No one knows, however, how they boost the immune response. In the March 19, 2013, issue of the Proceedings of the National Academy of Sciencesresearchers at National Jewish Health continue unraveling the mystery of adjuvants with a report that host DNA coats the alum adjuvant and induces two crucial cells to interact twice as long during the initial stimulation of the adaptive immune system.

“Alum makes T cells take a longer look at the antigen, which produces a better immune response,” said Philippa Marrack, PhD, senior author and professor of immunology at National Jewish Health. “Understanding how adjuvants work could help us make more effective vaccines. That is very important. Vaccines have saved millions of lives and been among the greatest advances in medical history.”

Live vaccines, containing weakened forms of an infectious organism, generally work fine by themselves. But vaccines containing dead organisms (inactivated vaccines) or pieces of the infectious organisms or their toxins (acellular or recombinant vaccines) generally need adjuvants to boost their effectiveness. Aluminum salts, known as alum, are the only adjuvant approved for use in the United States for routine preventive vaccines.

Adjuvants were first discovered as the result of empirical experiments with tetanus early in the 20th century. They have been widely used in many vaccines since the 1940s, including the Diphtheria/Tetanus/Pertussis (DtaP), Hepatitis, Haemophilus influenzae (Hib), typhoid and some flu vaccines. No one fully understands why adjuvants boost the effectiveness of nonliving vaccines.

Recently a Belgian team showed that DNA is involved in the adjuvant effect. When they administered a vaccine with adjuvant and DNase, an enzyme that digests DNA, the vaccine was less effective. The National Jewish Health team built on those findings to reveal the role that DNA plays.

The National Jewish Health team had previously shown that the process starts with a series of events similar to those that initiate responses to bacterial infections. Neutrophils, and other early responders in the immune system, flood into a site of potential infection, attack the foreign agent, in this case the alum vaccine, then quickly die in massive numbers.

Upon death the neutrophils release large amounts of DNA, which uncoils from its chromatin spools and acts somewhat like a net to entangle the foreign agent. Other cells then engulf the DNA-alum-vaccine complex. These antigen-presenting cells display small fragments of the vaccine on their surfaces for T-cells to recognize. T-cells drive the adaptive immune response, the one that recognizes and attacks the specific infectious agent, as opposed to the more general innate immune response.

T-cells are also the basis for effective vaccines. Some T-cells, and the B-cells stimulated by the T-cells, transform into memory cells once the infection has been cleared. Those memory cells help mount a quicker and stronger immune response if they see that organism again.

The National Jewish Health team showed that the DNA coating the adjuvant doubles the time that the T-cell engages the vaccine fragment on the surface of the antigen-presenting cell. When they added DNase to digest DNA, the T-cell engaged the vaccine fragment half as long, and the vaccine was less effective. Several of the findings were made possible by an innovative use of multi-photon microscopy to film the interaction of T-cells and antigen-presenting cells.

“The DNA makes the antigen-presenting cell stickier,” said Amy McKee, PhD, Instructor at the University of Colorado, and lead author of the paper. “We believe that extended engagement provides a stronger signal to the T-cell, which makes the immune response more robust.”

The researchers are not sure exactly what makes the antigen-presenting cell ‘stickier.’ When that an antigen-presenting cell engulfs free-floating DNA, the researchers believe it recognizes that something is amiss (DNA should not normally be floating around outside an intact cell nucleus) and becomes more activated. It may respond with an additional co-receptor to engage the T-cell or release a molecule that stimulates the T-cell. The researchers are now working to understand that process.

Finding genes for childhood obesity: Genome wide study identifies genetic variants associated with childhood obesity

Apr. 5, 2013 ? A new study has revealed promising targets for the development of new drugs against childhood obesity. Researchers have identified four genes newly associated with childhood obesity and an increased burden of rare genetic deletions and rearrangements in severely obese children. Gaining a better basic understanding of obesity will open new doors to clinically relevant research.

Researchers have identified four genes newly associated with severe childhood obesity. They also found an increased burden of rare structural variations in severely obese children.

The team found that structural variations can delete sections of DNA that help to maintain protein receptors known to be involved in the regulation of weight. These receptors are promising targets for the development of new drugs against obesity.

As one of the major health issues affecting modern societies, obesity has increasingly received public attention. Genes, behavior and environment, all contribute to the development of obesity.

Children with severe obesity are more likely to have a strong genetic contribution. This study has enhanced understanding of how both common and rare variants around specific genes and genetic regions are involved in severe childhood obesity.

?We?ve known for a long time that changes to our genes can increase our risk of obesity. For example, the gene FTO has been unequivocally associated with BMI, obesity and other obesity-related traits,? says Dr Eleanor Wheeler, first author from the Wellcome Trust Sanger Institute. ?In our study of severely obese children, we found that variations in or near two of the newly associated genes seem to have a comparable or greater effect on obesity than the FTO gene: PRKCH and RMST.?

The team found that different genes can be involved in severe childhood obesity compared to obesity in adults.

Rare genetic changes in one of the newly associated genes, LEPR, are known to cause a severe form of early onset obesity. The team identified a more common variant in this gene, found in 6 per cent of the population, that can increase a person?s risk of obesity. This finding is an example of where rare and more common variations around the same gene or region can influence the risk of severe obesity.

Some of the children in this study had an increased number of structural variations of their DNA that delete G-protein coupled receptors, important receptors in the regulation of weight. These receptors are key targets for current drug development and may have potential therapeutic implications for obesity.

?Some children will be obese because they have severe mutations, but our research indicates that some may have a combination of severe mutations and milder acting variants that in combination contribute to their obesity,? says Professor Sadaf Farooqi, co-lead author from the University of Cambridge. ?As we uncover more and more variants and genetic links, we will gain a better basic understanding of obesity, which in turn will open doors to areas of clinically relevant research.”

As part of the UK10K project (http://www.uk10k.org/) the team are now exploring all the genes of 1000 children with severe obesity in whom a diagnostic mutation has not been found. This work will find new severe mutations that may explain the causes of obesity in other children.

?Our study adds evidence that a range of both rare and common genetic variants are responsible for severe childhood obesity,? says Dr In?s Barroso, co-lead author from the Wellcome Trust Sanger Institute. ?This work brings us a step closer to understanding the biology underlying this severe form of childhood obesity and providing a potential diagnosis to the children and their parents.?

Stocks plunge after job growth disappoints

NEW YORK (AP) ? Stocks dropped sharply on Wall Street Friday after the government reported that U.S. employers added the fewest jobs in nine months in March and more people gave up looking for work. The report was worse than economists were expecting.

The Dow Jones industrial average fell 119 points to 14,485 as of 10:57 a.m. EDT. It was down as much as 171 points in the early going. It was the worst drop for the Dow since Feb. 25.

U.S. employers added 88,000 jobs in March, according to the Labor Department’s monthly survey. That’s half the pace of the previous six months, when an average of 196,000 jobs were added each month. It was a disappointment for markets following positive signs on housing and the job market over the winter.

The report, one of the most closely watched indicators of the economy, dented investors’ confidence that the U.S. was poised for a sustained recovery. The stock market has surged this year, pushing the Dow and the S&P 500 to record levels. The Dow closed at an all-time high on Tuesday and is still up 10 percent this year.

“Things are still looking decent, but there’s no doubt that this was a bit of disappointment,” said Brad Sorensen, Charles Schwab’s director of market and sector research. “We’re watching to see is this the start of another soft patch.”

In other trading, the Standard & Poor’s 500 index fell 14 points, or 0.9percent, to 1,545. All 10 industry groups in the index fell, led by a 1.5 percent drop in the information technology group. Cisco Systems fell 91 cents, or 4.4 percent, to $20.13. Oracle dropped 53 cents, or 1.6 percent, to $31.84.

Investors were reducing their exposure to risk. The industry that held up the best was utilities, with a decline of just 0.1 percent. The rich dividends and stable earnings provided by those companies make them attractive to investors who want to play it safe.

The yield on the 10-year Treasury note, which moves inversely to its price, plunged from 1.76 percent to 1.70 percent, its lowest level since December. The benchmark rate has declined sharply over the last month, from 2.06 percent on March 11, as demand for low-risk assets increased amid mounting evidence that growth in the U.S. economy is slowing.

The Nasdaq composite, which includes many technology companies, fell 37 points, or 1.2 percent, to 3,187. That’s worse than the declines of 0.8 percent in the Dow and 0.9 percent in the S&P.

F5 Networks, a network equipment and service provider based in Seattle, plunged 19 percent, the most of any S&P stock, after slashing its profit and revenue forecast. The company said its contract bookings fell sharply, as did its business with the federal government. The stock lost $17.28 to $73.14.

The S&P 500 is down 1.7 percent on the week and is on track for its biggest weekly loss since Nov. 9, when stocks fell after the Presidential election. Investors were worried that a divided government would fail to reach a budget agreement to avoid drastic tax hikes.

The Facebook Phone Is Smarter Than You Think

It’s really easy to want to hate a phone that is all Facebook all the time, especially when it gives the social network more and more of our data to mine for more ads to put on more prominent screens. Especially when the device itself, a middling (albeit sub-$100) HTC First?to wit, the first of many that will run Facebook’s Android-based “Home” software, CEO Mark Zuckerberg said at its launch event Thursday?is a a mediocre smartphone that doesn’t do anything exceptionally different. The so-called “Facebook Phone” runs a hidden version of Android; after the Facebook front door, it’s all the same.

So: Why would anyone want to get into this thing? The techies immediately suggested that people who love Facebook will flock to Home because it’s a simpler way to do just that?embrace something they already use on their phones all the time. But what if you’re just a casual Facebook browser, not obsessed with every last party photo and not quite part of a generation for whom Facebook messages pre-empt email? Well, there is more than a silver lining to Zuckerberg’s most blatant jump into your mobile life: This “Facebook Phone” offers some key user interface experiences that are way better than their counterparts on other Android phones?and even the iPhone.

These little things might not be enough to get you to hand over your data or eyeballs to Facebook, and certainly not to trade in your Apple device for some so-so Android. But the rest of the mobile industry would do well to learn from the truly brand-new things that entered the world today. Let’s take a moment to recognize the innovation unveiled Thursday from Menlo Park?even if it was from a guy in a hoodie and not a turtleneck:

Instead of having to open and close an application every time someone sends you an SMS or Facebook message, one of those little bubbles?hello up there, Sheryl!?pops up on the side of what you’re doing. Your bubble-headed friend’s chat shows up with a single tap. Or, you know, you can just ignore the head and keep on doing what you’re doing. It’s fast. It’s smooth. It’s more productive than disruptive. These are things you’re supposed to say about Apple. Plus if you’re chatting with someone and want to help each other, it provides a much faster way to share information, and on the go. And that’s something you’re supposed to be saying about Google. I haven’t tried the phone with my own two hands (launch day is April 12), but those on hand at Facebook HQ Thursday say Chat Heads is “responsive with very little lag” and not too intrusive. Once the heads pile up, you can swipe across to view the entire carousel.?

Notifications: Gone in a tap!

The new Facebook software has the same default notifications hub that comes on all Android devices?the pull down one that Apple stole from Google. But rather than having to get rid of those annoying updates from your LinkedIn app with a swipe, Facebook Home more prominently stacks the Facebook happenings you actually care about, and right on the home screen.

Now, that looks a lot similar to the notifications that pop up on any smart phone. But it’s very seamless?with or without Chat Heads. How many times have you swiped a notification to get rid of it on my iPhone, only to open some buggy app that never loads? With the Facebook phone, a simple tap rids the screen of an unwanted notification. You can also bundle them all up and dismiss them once and for all. But because it’s Facebook, you know, you might actually be interested.

Home’s homescreen: Now with a purpose.

So maybe Facebook photos aren’t the ideal thing to hover in the background of a phone. (We get enough baby pictures on the actual website, thanks.) The “Cover Feed,” as Facebook calls it, allows you to scroll through recently posted images from your Facebook feed, in full bleed and packed with information. That’s probably only a minor upgrade for people who constantly use their phone to look at photos on Facebook?or spend too many minutes in bed or on a bathroom line looking at Instagram.

But, imagine if we could put other, more useful things right there on the first stop, for quick access? As of right now, our phone backgrounds serve as high-tech time pieces, which seems a little dated.?

All of which is to say, it might not be worth it to get a Facebook phone?maybe for your teenage niece, or something. And it shouldn’t be forgotten that, after?the unveiling, Zuckerberg said advertising could be coming to the Cover Feed?and no other major phone is that invasive from the “on” button.?But no other major phone is this intuitive with the little things. So for a moment, let’s appreciate the finer parts of the Facebook phone.?

Review: Alfie Boe at Newcastle's Metro Radio Arena – Chronicle Live

Chronicle young reviewer Josh McLean reviews Alfie Boe sell-out Storyteller tour at Newcastle’s Metro Radio Arena

Alfie Boe returned to the North East with his sell-out Storyteller tour. Playing for over two hours, Alfie certainly gave his fans what they wanted ? a night of pure entertainment.

The opening act was Emilia Mitiku, a sultry Swedish artist with a vintage pop feel. She had beautiful melodies and great vocals.

Alfie entered the stage to a loud applause and had the crowd clapping along to songs in seconds.

Performing in a half-size arena, the atmosphere was intimate which made the whole audience feel included.

Throughout the night Alfie was running, jumping and dancing his way through his impressive repertoire ? still managing to hit those high notes as he shot from one side of the stage to the next.

Performing The First Time Ever I Saw Your Face, there was a real stillness in the arena as everyone felt the passion and emotion that Alfie creates with his majestic voice. Another cover soon followed, this time Simon and Garfunkel?s Bridge over Troubled Water and the whole crowd was in awe.

He then took time to talk about being back in Newcastle and how he enjoyed the feel of the city. At this point, sofas had been placed on stage that allowed the setting of an acoustic segment to perform some of his classical pieces.

Having gained a following from his run in the musical phenomenon Les Miserables, it was clear to see those fans were in the audience.

At the mention of the words, the audience was screaming with excitement. Performing Bring Him Home, he received his first of many standing ovations. It was incredible and the highlight of the night.?

By this point, Alfie had the audience in the palm of his hand. He encouraged his legion of fans to come stand at the front of the stage, where he shook their hands and kissed people on the cheek. He looked more like a rock god than classical tenor.

Towards the end of the show he ran through crowds, climbing over speakers and up each set of stairs to meet all his fans. Not even security could stop him as they chased after him.

He was having fun and so was the audience.

The night closed with an encore that was desperately wanted. He had planned to perform a track with himself on guitar, but after a slight technical glitch, he performed it a capella style. It was with out a doubt better than what he could have originally intended.

Alfie was faultless throughout, his stunning vocals were fantastic to hear and he was a pleasure to watch.

Report: Saudi Arabia may try to end anonymity for Twitter users

Saudi Arabia may try to end anonymity for Twitter users in the country by limiting access to the site to people who register their identification documents, the Arab News daily reported on Saturday.

Last week, local media reported the government had asked telecom companies to look at ways they could monitor, or block, free internet phone services such as Skype.

Twitter is highly popular with Saudis and has stirred broad debate on subjects ranging from religion to politics in a country where such public discussion had been considered at best unseemly and sometimes illegal.

Early this month, the security spokesman for Saudi Arabia’s Interior Ministry described social networking, particularly Twitter, as a tool used by militants to stir social unrest.

The country’s Grand Mufti, Saudi Arabia’s top cleric, last week described users of the microblogging site as “clowns” wasting time with frivolous and even harmful discussions, local newspapers reported.

“A source at (the regulator) described the move as a natural result of the successful implementation of (its) decision to add a user’s identification numbers while topping up mobile phone credit,” Arab News reported.

That would not necessarily make a user’s identity visible to other users of the site, but it would mean the Saudi government could monitor the tweets of individual Saudis.

The English-language daily and sister paper to the Saudi-owned pan-Arab Asharq al-Awsat newspaper did not explain how the authorities might be able to restrict ability to post on Twitter. Both newspapers belong to a publishing group owned by the ruling family and run by a son of Crown Prince Salman.

Internet service providers are legally obliged to block websites showing content deemed pornographic.

One of the big investors in Twitter is Saudi Arabian billionaire Prince Alwaleed bin Talal, a nephew of King Abdullah who also holds significant stakes in Citi Group, News Corp and Apple through his Kingdom Holding Company.

The country’s telecom regulator, Communications and Information Technology Commission (CITC) did not immediately responded to requests for comment on the report. Last week it did not comment on the report it was seeking to restrict Skype use.

A spokeswoman for Kingdom Holding said Prince Alwaleed was not available to comment.

“There are people who misuse the social networking and try to send false information and false evaluation of the situation in the kingdom and the way the policemen in the kingdom are dealing with these situations,” said Major General Mansour Turki, the security spokesman, at a news conference on Mar 8.

At a separate interview with Reuters this month, Turki argued that a small number of supporters of al Qaeda and activists from Saudi Arabia’s Shi’ite minority used social media to stir wider sympathy for their goals and social unrest.

However, he also argued against banning the site.

Two weeks ago one of Saudi Arabia’s most prominent clerics, Salman al-Awdah, who has 2.4 million followers on the site, used Twitter to attack the government’s security policy as too harsh and call for better services. He warned it might otherwise face “the spark of violence”.

Two leading Saudi human rights activists were sentenced to long prison terms this month for a variety of offences including “internet crimes” because they had used Twitter and other sites to attack the government.

Some top princes in the monarchy now use Twitter themselves and Crown Prince Salman, King Abdullah’s designated heir and also Defence Minister, recently opened an official account.

FRIDAY, March 29 (HealthDay News) ? As more patients undergo a minimally invasive procedure to remove kidney stones, the rate of complications from the surgery is also rising, according to a new study.

The procedure ? called percutaneous nephrolithotomy (PCNL) ? involves making a small incision in the back and using a hollow scope to remove medium to large kidney stones.

Although the death rate related to the procedure remained low over the 10-year study period, certain complications, including blood infection, have soared. Patients were at higher risk of developing complications if they were older, sicker and treated in more recent years, the study found.

?We believe the broad use of this procedure, especially in older and sicker patients, may be the reason [for the increased rate of complications],? Dr. Khurshid Ghani and colleagues said in a news release from Henry Ford Hospital in Detroit.

For the study, published recently in the Journal of Urology, the researchers analyzed data from more than 80,000 patients over age 18 in the United States who had the procedure between 1999 and 2009.

During that time, its use increased 47 percent, most notably among women.

The presence of other disorders or diseases at the time of surgery increased during the study period, and overall complications rose from about 12 percent to nearly 16 percent. Of particular note, the incidence of blood infection (sepsis) doubled from 1.2 percent to 2.4 percent.

The rate of death related to the procedure remained essentially unchanged at 0 to 0.4 percent. Deaths that did occur were in older patients, the study found.
